Free erythrocyte protoporphyrin.

چکیده
Porphyrins are widely distributed in living cells throughout nature and play essential roles in various metabolic processes such as photosynthesis, transportation of oxygen and cellular respiration. In man the most important of the porphyrins is protoporphyrin 9, type III, which in combination with iron and specific proteins forms such compounds as hemoglobin, myoglobin, cytochrome, peroxidase and catalase. In addition to its occurrence in these compounds, protoporphyrin has been found in apparently uncombined or free form in erythrocytes, hence the term "free erythrocyte protoporphyrin" (1-3). Cartwright and associates (4) recently have summarized many of the salient features of previous work concerning free erythrocyte protoporphyrin. Suffice it to say here that the origin, function and fate of free protoporphyrin in erythrocytes remain uncertain. Indirect evidence suggests that it serves as an intermediary in synthesis of hemoglobin. Increased free erythrocyte protoporphyrin is thought to result from an interruption in normal synthesis of hemoglobin, as in the presence of iron deficiency states, increased erythropoiesis, or a disturbance of iron metabolism (5). Decreased levels have been ascribed to a deficiency in synthesis of protoporphyrin (6). There is evidence that reticulocytes contain more free protoporphyrin than do mature erythrocytes (7, 8). This investigation has been concerned with the study of amounts of free protoporphyrin 2 in eryth-

EFFECT OF CHRONIC LEAD POISONING ON ERYTHROCYTE PROTOPORPHYRIN IN RATS
Research has shown that exposure to lead may have adverse effects at different blood lead concentrations. Lead inhibits at least two enzymes that are essential for the formation of heme, and because of the interaction of lead with these enzymes, no iron is inserted into protoporphyrin. Therefore the concentration of protoporphyrin increases in erythrocytes. The concentration of lead was mea...
متن کاملPitfalls in Erythrocyte Protoporphyrin Measurement for Diagnosis and Monitoring of Protoporphyrias.
BACKGROUND Laboratory diagnosis of erythropoietic protoporphyria (EPP) requires a marked increase in total erythrocyte protoporphyrin (300-5000 μg/dL erythrocytes, reference interval <80 μg/dL) and a predominance (85%-100%) of metal-free protoporphyrin [normal, mostly zinc protoporphyrin (reference intervals for the zinc protoporphyrin proportion have not been established)]; plasma porphyrins a...
متن کاملSideroblastic anemia with markedly increased free erythrocyte protoporphyrin without dermal photosensitivity.
A patient with idiopathic sideroblastic anemia and atypical clinical and biochemical findings is described. He had a greatly increased erythrocyte and plasma protoporphyrin, but normal urine and fecal porphyrins. The erythrocyte protoporphyrin had a fluorescence spectrum typical of free protoporphyrin, but caused no photosensitivity. Bone marrow metal chelatase activity was normal. There were n...
متن کاملFree erythrocyte protoporphyrin level and nerve conduction velocity in end-stage renal disease.
Increased free erythrocyte protoporphyrin concentrations and depressed motor nerve conduction velocities (MNCV) were observed in 45 patients on maintenance haemodialysis. Neither of these findings could be correlated with age, duration or frequency of dialysis, or the degree of uraemia present. A strong negative correlation (r=--0-53; P less than 0-001), however, existed between the free erythr...
متن کاملLight-induced release of protoporphyrin, but not of zinc protoporphyrin, from erythrocytes in a patient with greatly elevated erythrocyte protoporphyrin.
A patient with greatly increased erythrocyte protoporphyrin, but normal porphyrins in urine and feces, is described. The patient later developed a malignant lymphoma, and the reason why she accumulated protoporphyrin in her erythrocytes is not known. The protoporphyrin in the erythrocytes consisted of two types of protoporphyrin, free protoporphyrin (30%) and zinc protoporphyrin (70%). Upon irr...
